基因PTEN(phosphatase and tensin homology deleted on chromosome ten )属于抑癌基因,是一个重要的肿瘤抑制因子,在细胞增殖、存活以及细胞代谢活动中发挥关键作用,主要通过PI3K/AKT信号通路发挥作用。PTEN的失活或表达下调与多种癌症的发生和发展密切相关,PTEN的失活通常会导致PI3K/AKT信号通路的过度激活,进而促进细胞增殖、生存和迁移,这可能通过调控下游的mTOR、FOXO和GSK-3β等信号通路实现,PTEN表达水平的下降通常与食管癌患者的不良预后相关,因此,PTEN表达水平可作为食管癌患者预后的潜在生物标志物,有助于疾病的早期诊断和个性化治疗策略的制定。由于PTEN在食管癌发生发展中的重要作用,靶向PTEN/PI3K/AKT信号通路成为一种潜在的治疗策略。尽管PTEN的失活与食管癌的恶性行为和不良预后密切相关,但针对PTEN/PI3K/AKT通路的靶向治疗仍需要进一步研究和临床验证,未来,深入了解PTEN在食管癌中的分子机制和寻找有效的靶向干预措施将有助于改善食管癌患者的治疗效果和生存率,在此作一综述,总结PTEN在食管癌中的研究现状,为之后更进一步研究PTEN与食管癌的相关性奠定基础。
