目的:本文旨在研究死盒解旋酶33(DHX33)在调控肺癌细胞迁移和凋亡中的作用。方法:利用shRNA技术敲低肺癌细胞NCI-H2228中的DHX33,并通过实时荧光定量PCR和Western blotting验证DHX33的敲低效率;通过细胞划痕和流式细胞术检测DHX33敲低后对肺癌细胞迁移和凋亡的影响。结果:利用shRNA技术在mRNA水平(P<0.0001)和蛋白水平均显著抑制了肺癌细胞中DHX33的表达量。与对照组肺癌细胞相比,DHX33的敲低显著降低了肺癌细胞的迁移能力(P<0.05),并且还显著促进肺癌细胞的凋亡水平(P<0.05)。结论:敲低DHX33可抑制肺癌细胞NCI-H2228的迁移并促进其凋亡。
