近年来,肥胖已经成为全球重大公共卫生问题。肥胖显著增加2型糖尿病、心血管疾病等慢性疾病的发病风险,给医疗体系带来了巨大的经济压力。从糖脂能量代谢视角,梳理了肥胖发病的关键分子信号通路,肥胖通过中枢-外周“失耦合”形成“高摄入-低消耗-激素抵抗-炎症-脂质沉积”恶性循环,导致胰岛素抵抗和2型糖尿病。深入阐明肥胖发病的分子信号网络,以期为开发新型防治策略提供理论依据,突破当前肥胖治疗的瓶颈。
