目的:探究鹿茸抗激素性股骨头坏死大鼠股骨头成骨细胞凋亡的作用及机制。方法:选择3月龄健康雌性SD大鼠30只,体质量(200±20)g,随机分为鹿茸组及对照组。常规喂养1周后,分别予醋酸泼尼松(3.5 mg/kg)、鹿茸(70 mg/kg)+ 醋酸泼尼松(3.5 mg/kg)灌胃13 周,进行股骨头Micro CT扫描及重建、骨形态计量学分析,通过ELISA检测成骨分化相关基因(Osteriex和RUNX2),TUNEL法检测股骨头成骨细胞凋亡情况,分离两组成骨细胞外泌体,检测外泌体中成骨分化相关基因(Osteriex和RUNX2)表达情况,将外泌体与成骨细胞联合培养,利用CCK-8法检测细胞增殖,流式细胞术检测细胞凋亡。结果:鹿茸组骨小梁结构破坏程度明显优于对照组,成骨细胞及外泌体中成骨分化相关基因(Osteriex和RUNX2)表达鹿茸组显著高于对照组(P<0.05),鹿茸组成骨细胞凋亡率降低(P<0.05),外泌体与成骨细胞联合培养中鹿茸组相比于对照组,成骨细胞增长率较高,细胞凋亡较低(P<0.05)。结论:鹿茸可能通过干预成骨细胞外泌体发挥抗成骨细胞凋亡从而发挥治疗大鼠SANFH的作用。
